汉防己碱滴眼液对慢性高眼压兔模型治疗作用的研究

摘要

目的 探讨汉防己碱滴眼液对慢性高眼压兔模型的治疗作用.方法 新西兰兔18只,随机分为空白组、模型组及给药组,每组6只.利用复方卡波姆溶液进行慢性高眼压造模,给药组家兔给予汉防己碱滴眼液(浓度0.1g·L-1)滴眼,模型组及空白组则给予等量生理盐水滴眼.治疗7d后处死各组家兔,取视网膜,分为2份,并分别用Thy-1染色及制备匀浆.利用光镜观测Thy-1染色并对其视网膜神经节细胞(RGCs)进行计数;检测匀浆液中谷胱甘肽过氧化物酶(GSH-PX)、超氧化物歧化酶(SOD)及丙二醛(MDA)含量.结果 空白组,给药组与模型组RGCs计数分别为每高倍视野(21.2±1.7),(20.1±2.5)及(15.2±2.3)个,空白组与给药组分别与模型组比较,差异有统计学意义(P＜0.05);模型组中内丛状层变薄,外核层细胞排列紊乱、数目减少且间隙扩大;给药组及空白组家兔视网膜各层组织层次分明、结构清晰且细胞排列整齐.给药组及空白组家兔视网膜匀浆液中SOD,MDA及GSH-PX水平均明显优于模型组.结论 汉防己碱滴眼液对慢性高眼压兔模型具有较好的治疗作用.%Objective To explore the protective effect of tetrandrine on the chronic ocular hypertension in rabbits.Methods Eighteen rabbits were randomized into 3 groups:blank control group,model group and treatment group.Carbomer solution was injected to make the chronic ocular hypertension model in the model group and treatment group.Tetrandrine eye drops (0.1 g · L-1) were adopted to treatment group while isometric saline was used in model group and blank control group.Seven days after treatment,the rabbits were killed and the retinas were obtained and assigned into two parts.One part was dyed by Thy-1 and the other was homogenized.The RGCs counts and the pathologic changes were recorded.SOD,MDA,and GSH-PX levels were observed.Results The RGCs counts of blank control group,model group and treatment group were (21.2±1.7),(20.1±2.5) and (15.2±2.3),respectively.The promotion of SOD,MDA and GSH-PX levels in the blank control group and treatment group were more excellent than the model group.Conclusions Tetrandrine has a good treatment effect on the chronic ocular hypertension in rabbits.
