18 F-氟赤硝基咪唑micro PET/CT评价裸鼠乳腺癌早期放疗疗效实验研究

         

摘要

目的 探讨18 F-氟赤硝基咪唑micro PET/CT 评价裸鼠MDA-MB231 乳腺癌早期放疗疗效的价值.方法 建立 16 只裸鼠MDA-MB231 乳腺癌模型,将其按照随机对照原则分为两组:对照组(A 组)、放疗组(B 组),每组8 只.每组裸鼠行micro PET/CT 显像,测定每只裸鼠肿瘤SUVmax 值.完成显像后,常规HE 染色观察每组肿瘤组织形态学特征,免疫组化方法测定每组肿瘤细胞乏氧诱导因子-1α(HIF-1α)的表达情况.结果 放疗前,对照组与放疗组SUVmax 值差异无统计学意义(t = 0. 375,P > 0. 05).放疗组放疗后48 h裸鼠肿瘤组织SUVmax 值较放疗前(t = 9. 958,P 0. 05). The SUVmax value of tumor tissue when 48 h after radiotherapy was significantly lower than that before radiotherapy (t = 9. 958,P < 0. 05) and 24 h after radiotherapy(t = 16. 506,P < 0. 05),the difference was statistically significant(F = 58. 860,P < 0. 05). The SUVmax value of tumor tissue when 48 h after radiotherapy was also lower than before radiotherapy(t = 5. 405,P < 0. 05). More obvious necrosis was observed in the radiotherapy group than the control group by HE staining. The expressions of HIF-1α in the radiotherapy group were significantly decreased compared with the control group(t = 14. 802,P < 0. 05). The tumor SUVmax value was positively correlated with HIF-1α expression(r = 0. 865,P < 0. 05). Conclusion 18F-FETNIM micro PET /CT not only can be used to detect the hypoxic state within the tumor,but also can evaluate the early radiotherapeutic efficacy in MDA-MB231 breast cancer in nude mice.
