Glucose marked by 3-amino-9-ethylcarbazole ( AEC) was sprayed onto tobacco leaf and observed with Leica-DMLB fluorescence microscope to evaluate casing absorption by tobacco leaf. Results showed that glucose failed to penetrate tissue after 8 h under 60°C , 70% RH, other than adhered to surface. Further observation with samples sprayed onto cut slices suggested that cuticle of tobacco leafs prevented glucose molecule from penetrating.%为评价料液在烟叶中的吸收效果,采用3-氨基-9-乙基咔唑(AEC)标记葡萄糖为标记物,通过Leica-DMLB荧光显微镜对葡萄糖在烟叶中的渗透效果进行观测.结果表明喷洒AEC标记葡萄糖的烟叶经过60℃、70%相对湿度条件下渗透8h,低温冰冻切片,观察发现所施加葡萄糖均附着在烟叶表面,未能渗透到烟叶组织内部.进一步观察切片后喷洒AEC标记葡萄糖的烟叶,认为叶片表面的角质层阻挡了葡萄糖分子的渗入.
