Objective To investigate the effect of ferulic acid on ventricular collagen remodeling in mice with pressure over load induced chronic heart failure. Methods A total of 40 wild type C57BL/6 male mice were divided into sham group(Sham) , sham with ferulic acid group (SFA) ,TAC group(TAC) and TAC with ferulic acid group (TFA). After 8 weeks, echocardio graphy was performed to test the morphology and function of the heart. PSR. staining was used to detect the collagen deposition. RT PCR and immunohistochemistry were used to detect the expression of mRNA and protein of MMP9 in different groups. Results TFA exhibited fewer indications of cardiac dysfunction(including increases in LVFS and LVEF,and decreases in LVEDD and LVESD)than the TAC. In comparison to TAC,the TFA attenuated the pressure overload induced changes in the expression of MMP9 both in mRNA and protein levels. The augmentation in cardiac fibrosis area observed after PSR staining in the TFA was also markedly attenuated in comparison to that in TAC. Conclusion Ferulic acid can markedly reduce the collagen in mice with chronic heart failure by blocking MMP9.%目的 探讨阿魏酸对慢性心力衰竭小鼠心室胶原重构的影响.方法 40只小鼠分为假手术组、假手术阿魏酸组、心衰组、心衰阿魏酸组;心衰组和心衰阿魏酸组采用主动脉缩窄术制作压力超负荷慢性心力衰竭小鼠模型.手术8周后行超声心动图检测心脏结构和功能变化;心肌天狼猩红染色观察胶原变化;取左室游离壁心室肌,利用RT-PCR及免疫组化方法观察心肌纤维化指标基质金属蛋白酶9(MMP9)mRNA和蛋白表达变化.结果 与假手术组相比,心衰组、心衰阿魏酸组左室内径增大、室壁增厚,射血分数降低(均P<0.05),MMP9 mRNA和蛋白表达均增强,胶原面积占总面积的比值明显增大(均P<0.05);与心衰组相比,心衰阿魏酸组左室内径减小、室壁肥厚程度降低,射血分数升高(均P<0.05),MMP9 mRNA和蛋白表达水平减弱,胶原面积明显减小.结论 阿魏酸能够抑制慢性心力衰竭小鼠心肌MMP9的表达,在一定程度上改善了心室胶原重构和心脏功能.
