高粱Pht1基因家族的鉴定及其进化和表达分析

         

摘要

磷是生物大分子的重要组成成分,也是植物生长所必需的重要营养元素.利用Phytozome数据库,通过拟南芥Pht1家族、水稻OsPht1家族基因序列在高粱全基因组中的同源比对,鉴定并获得了11个高粱Pht1家族成员的基因序列、基因结构、蛋白序列和染色体定位等信息.利用荧光定量的方法,检测了11个SbPht1基因在缺磷和高磷处理下的表达差异.结果表明,在缺磷条件下,大部分SbPht1基因表达量上升.高磷胁迫下,SbPT1、SbPT4、SbPT6、SbPT7、SbPT8、SbPT9和SbPT10在根中的表达量上升,SbPT3和SbPT5在地上部的表达量上升.分析高粱全基因组磷转运蛋白,对于研究植物在磷吸收过程中的分子生物学基础,改良植物吸收和利用磷素意义重大.%Phosphorus is a significant component of biological molecules, and it is also an important nutrient for plant growth. To identify putative Pht1 family genes in S. bicolor, we performed BLAST searches against the S. bicolor ge-nome database Phytozome using the well-characterized Arabidopsis AtPht and rice OsPht1 family as queries, and 11 non-redundant putative SbPht1 genes were identified. Information of gene sequence, gene structure, protein se-quence and chromosomal location were downloaded from Phytozome database. Quantitative real-time polymerase chain reaction ( qRT-PCR) was used to analyze the expression patterns of 11 SbPht1 genes under phosphorus defi-ciency and high phosphorus treatment. The results showed that most of the SbPht1 genes expression was increased under the condition of phosphorus deficiency. Under high phosphorus treatment, the expression of SbPT1, SbPT4, SbPT6, SbPT7, SbPT8, SbPT9 and SbPT10 was up-regulated in the roots. In the shoots, the expression of SbPT3 and SbPT5 was increased. Through the analysis of phosphate transport protein from the whole genome of sorghum, this paper provides the molecular biological basis for the study of phosphorus, and has a great significance in the im-provement of plant absorption and utilization of phosphorus.
