红叶石楠组培苗玻璃化影响因子及其克服技术研究

         

摘要

Factors in red tip Photinia (Photinia fraseri) vitrification in vitro were studied. The results showed that rich mass elements of mineral nutrients or high dosage of 6 - BA in the media increased the rate of vitrified plantlets, lower temperature or higher concentration of gelling agents reduced the rate compared with normal culture condition. More vitrified plantlets were found in the media solidified by carrageenan than in the media by agar in the similar gelling degree. The vitrification rate went down with the increase of multiplication generations. The plantlets with some leaves vitrified could be de - vitrified at 61. 67% ,57. 92% and 47. 08% respectively, treated in the way as follows for 60 days: 1. culture temperature changed from 25℃ all daytime to 25℃ for 16 hours in light period and 20℃ for 8 hours in dark period, 2. concentration of 6 - BA decreased from 1.5 mg/L to 0.5 mg/L, 3. the quantity of solidifying agent Carageenan added to the media increased from 7.0 g/L to 8.0 g/L, but the shoots vitrified completely had no change.%通过改良MS+6-BA无菌培养红叶石楠,结果表明:培养基离子浓度或细胞分裂素浓度偏高,组培苗玻璃化的比例均增加,降低培养温度或提高培养基硬度玻璃化苗均减少,相近凝固效果的卡拉胶比琼脂易导致玻璃化苗;繁殖代数增加,玻璃化比例降低.培养温度由恒温改为变温、6-BA浓度由1.5 mg/L降为0.5 mg/L、卡拉胶由7.0 g/L增加到8.0 g/L,培养60 d后分别能将61.67%、57.92%和47.08%的仅部分叶片出现玻璃化的红叶石楠组培苗转为正常,部分叶片玻璃化如培养条件不变,或茎、叶完全玻璃化,即使改变培养条件,均不能转变.
