储藏条件和种子粒径对天师栗种子生活力的影响研究

         

摘要

通过探索天师栗种子的适宜储藏条件及播种种子粒径,以提高种子生活力,节约种子繁殖过程中种子用量.采用二因素多水平试验设计,筛选TTC染色法测定天师栗种子生活力的最佳TTC浓度和染色时间,同时运用TTC染色法对不同储藏条件下不同粒径的天师栗种子生活力进行比较研究.结果表明,TTC染色法测定天师栗种子生活力的最佳条件为:0.20%浓度的TTC溶液染色11~13 h;沙藏方式储藏的种子生活力较其它方式均高,达85.30%,并且差异达到显著水平;大粒径种子(φ≥4.5 cm)和中粒径种子(3.0 cm≤φ<4.5 cm)的生活力比小粒径种子(φ<3.0 cm)生活力高.故在天师栗种子繁殖过程中,宜采用沙藏方式储藏,并选用较大粒径种子(φ≥3.0 cm).%In order to improve the seed viability of Aesculus wilsonii and to save its seed usage during seed propagation, the author studied the optimum conditions for its storage and the suitable seed size for its sowing through adopting two-factor and multi-level experiment design.The best TTC concentration and dyeing time for the determination of seed viability of A.wilsonii by using TTC method were screened, and the viabilities of different sizes of A.wilsonii seeds under various storage conditions were compared.The results showed that the optimal determination condition for the seed viability of A.wilsonii was staining for 11~13 h with 0.2% TTC solution.The viability of A.wilsonii seeds under sand-storage condition was the highest (85.30%), and it was significantly higher than that under other storage conditions.The large size of seeds (grain diameter φ≥4.5 cm) and middle size of seeds (3.0 cm≤φ<4.5 cm) had a higher viability than the small size of seeds (φ<3.0 cm).Therefore, in the process of seed propagation of A.wilsonii, the larger size of seeds (φ≥3.0 cm) should be used and they should be stored in sand.
