γ射线辐照血小板不同保存时间质量参数变化分析

         

摘要

目的 分析经γ射线辐照前后手工浓缩血小板和单采血小板在不同保存时间的CD62P、平均血小板体积(MPv)、分布宽度(PI)W)的变化.方法 随机选择手工浓缩血小板(PCs)和单采血小板(A-PCs)各20袋,每袋平均分为两份,其中一份经137铯辐照.剂量25Gy,另一份不辐照;在22℃±2℃的血小板恒温振荡保存箱内保存72h.经辐照的手工血小板和单采血小板分别设为观察组1、观察组2;不辐照的血小板分别设为对照组1、对照组2.流式细胞仪检测血小板辐照前及保存24h、72h后P选择素表达情况,全自动血球计数仪检测MPV、PDW.结果 辐照组与对照组血小板表达CD62P百分率均随保存时间延长而增高,保存72h后两组差异有统计学意义(P<0.01、P<0.05);辐照组与对照组的血小板随着保存时间的延长MPv、PDW逐渐增加,但相同时间段相比差异无统计学意义(P>0.05).结论 γ射线照射对血小板的质量无明显影响,但辐照后血小板应尽可能在72h内应用.%Objective To analyze the changes of CD62P, mean platelet volume(MPV), and platelet distribution width(PDW) at different preservation time points after gamma irradiation. Methods Twenty bags of concentrated platelets and 20 bags of apheresis platelets were randomly selected. The platelets in each bag were divided into two parts. One part was irradiated with 137 cesium at 25Gy and the other part was not irradiated. The two parts were preserved at 22℃±2℃ for 72h. The concentrated platelets and apheresis platelets were considered as irradiation groups 1 and 2, the platelets not irradiated were considered as control groups 1 and 2. Expression of CD62P in platelets was detected by flow cytometry before and 24 and 72h after irradiation, respectively. MPV and PDW were calculated with a blood counter. Results The expression rate of CD62P in platelets increased with the preservation time of platelets in both irradiation and control groups with a significant difference 72h after preservation between the two groups (P<0.05). The MPV and PDW also increased with the preservation time of platelets in both irradiation and control groups with no significant difference between the two groups(P>0.05). Conclusion Platelets should be used within 72h after irradiation although Gamma irradiation has no significant effect on them.
