Background: Carcinoma of the urinary bladder occurs in highfrequency in china where it is the lOth most common cancer, accountingfor 17,365 deaths in 2005, and mortality has steadily increased between1991 and 2005. Growth of solid tumors and the formation of metastases aredependent on both the degree of tumor vascularization and the rate ofangiogenesis. Specific binding of inducers to cellular ligands such asVEGF on endothelial cells triggers the sequence of events leading to theformation of new vessels. The potency to induce angiogenesis is aproperty that has excited the current interest in VEGF. Thus, the potentialof VEGF as a new prognostic factor and target for anti cancer therapy inhuman bladder cancer at the VEGF protein level was studied. Vascular endothelial growth factor (VEGF) regulatesneovascularization in malignant cells. VEGF as a mitogen is thought toalter cancer cell formation and tumor. Objectives: To study vascular endothelial growth factor (VEGF) inurinary bladder carcinoma and its relationship with pathological stageand histological grade progression; and to evaluate theirhistopathological relevance in transitional cell carcinoma (TCC) ofurinary bladder. Method: Tissue samples from 13 patients with urinary bladdercancer were examined with an immunohistochemical stain for theexpression of the VEGF gene. The expression rate was compared t0 4specimens of normal bladder mucosae were thinly dissected from thetotal cystectomy specimens who had proved negative for carcinoma. Therelationship between VEGF protein expression and clinicopathological parameters finding was statistically analyzed. The relevance of VEGFexpression in cancer formation and it growth was also observed. The results revealed significant differences between normalurothelium (O%) and cancer tissue (69.2%) for the positive staining ofVEGF protein (P<0.05). With the progression of tumor grade and clinicalstaging, the positive rate of VEGF gene expression significantlyincreased. Expression of the VEGF gene in the invasive group wasgreater than that in the noninvasive group and significantly different Conclusion: The results revealed that expression of the VEGF geneis proportional to the formation and progression of TCC. Therefore,abnormal expression of VEGF genes can be used as a prognostic markerin TCC of urinary bladder.
