MIR-210 UP-REGULATES VEGF SIGNALING FOR ANGIOGENESIS IN VITRO

MicroRNAs(miRNA) are short non-coding RNA molecules that negatively regulate gene expression by posttranslational suppression or target mRNA degradation in a sequence-specific manner. In the present study, we investigated miRNA role in regulating angiogenesis in vitro and explored the underlying mechanisms involved in these events, we found that overexpression of miR-210 in human umbilical vein endothelial cells (HUVE-12) by molecular cloning technology stimulated the formation of capillary-like structures on Matrigel . To elucidate the further mechanisms of miR-210 regultion of angiogenesis, we analyzed the expression of VEGF /VEGFR2 at both mRNA and protein levels in HUVEC-12 cell line after miR210 overexpression. The rsults showed that VEGF/VEGFR2 expression in cells overexpression miR210 were markedly up-regulated compared with that in control. It suggested that VEGF signaling pathway may be involved in the regulation mechanism of miR-210mediated angiogenesis.
