@@Antitumor therapy has always been a great challenge to the scientists. The problems of traditional chemotherapeutics include the unspecific distribution, poor activity, serious toxicities as well as poor water solubility of some anticancer agents. Although PEGylation of drug delivery system is able to increase drug accumulation in tumor tissue, it does not enhance the intracellular delivery of drug, which greatly limits the efficacy of antitumor drugs. Therefore it seems more important to increase the specific interaction of drugs with tumor cells. Tumor growth, invasion and metastasis are all angiogenesis dependent. Tumor mass over than 2 mm3 is dependent on angiogenesis to supply nutrients and oxygen. Angiogenesis also provide the chances of the hematogenous diffusion and distant metastasis of tumor. Block and inhibition of tumor angiogenesis, therefore cutting off the supply of nutrients and oxygen, may be an effect strategy for tumor therapy. Tumor vasculature-targeted therapy has become an important component of tumor treatment.
