Preparation of puerarin microemulsion and its pharmacokinetics in mice

Aim To improve the oral bioavailabilty of puerarin, to study preparation of puerarin microemulsion. To study its physicochemical properties, its pharmacokinetical characteristics. MethodThe prescriptions is determined by pseudo -ternary phase diagrams. The tests of physicochemical properties including viscosity, conductivity, partical size, stability and etc. Compared with puerarin suspesion, the concentration of puerarin in serum of mice and their phamacokinetic behaviors after oral administration were studied by FIPLC. The phamacokinetic parameters were computed by software program 3P97. Results The concentration was 65mg·ml-1, with particle size of 40.2±5.9nm. Puerarin microemulsion showed a slower, longer serum concentration-time profile, as compared with suspension oral administration in both absorption and elimination phase. The bioavailability of puerarin microemulsion was 37.91%. Conclusion Particles of Puerarin microemulsion were round and homogeneous. Puerarin microemulsion showed good stabilit. Puerarin microemulsion dramatically enhance bioavailability of puerarin after oral administration.
