Regulation is a key challenge of design in Synthetic Biology. [1] In the past, inducible promoters have formed the regulatory backbone of some of the field's most successful designs.[2-4] However, only a limited number of inducible promoters are commonly used and new regulatory tools are needed to enable the creation of more complex systems.[5-7] One method to supplement wild-type inducible promoters is to build so-called synthetic inducible promoters (SIPs). These SIPs have shown potential as rationally designed regulatory parts.[8, 9]
