We created two libraries of SIPs from the Anderson constitutive promoter set, controlled by the XylR and LuxR transcription factors. Patterns between constitutive and SIP strength were observed, confirming the potential for rational design. Using transcriptional and translational predictive modelling, an SIP was inserted into a biological AND gate. This improved the system, demonstrating the potential of SIPs for the rational design of synthetic biology devices.
