Enhanced Antitumor Effects of Gemcitabine-loaded Temperature-Sensitive Liposomes by Hyperthermia

New temperature-sensitive liposomes (TSL) that could release their contents by controlling the temperature were prepared and studied for chemotherapeutic purposes to enhance the release of anticancer drugs at tumor sites. The effectiveness of the combination of gemcitabine (Gem), gemcitabine-loaded TSL (G-TSL) and hyperthermia (HT) is described. In vitro drug release from TSL was significantly increased by elevated temperature compared with non temperature-sensitive liposomes (NTSL). To investigate the therapeutic efficacy of G-TSL, the in vivo antitumor activity was evaluated in CT-26 murine colon adenocarcinoma tumorbearing mice in combination with HT. G-TSL inhibited tumor growth to a much greater extent than the control or free drug. Moreover, the antitumor activity of G-TSL was enhanced significantly when they were administered in combination with HT. GTSL were found to be highly efficacious carriers for the in vivo delivery of anticancer drugs and to have potential anticancer applications in combination with HT.
