Intracellular DNA Release and Elimination Correlate Poorly with the Transgene Expression after NonViral Transfection

A novel method based on real time quantitative RTPCR (qRTPCR) and Dnase treatment was developed to study the intracellular DNA elimination and release kinetics after nonviral transfection. Overall, neither total nor released amount of intracellular DNA correlated with transfection efficiency. This suggests that localization of DNA in active form especially at the nuclear level may determinate transfection efficiency.
