Rapid disintegrating lyophilised tablets were formulated using naturally occurring amino acids including alanine, histidine, arginine and methionine at varied concentration. Freeze-drying of the formulations produced dry and elegant tablets, which were characterised for their mechanical property, porosity and disintegration time. The disintegration mechanisms were also elucidated. The results showed that inclusion of the amino acids within the tablet matrix improved the mechanical property. However, only alanine and histidine were able to accelerate the disintegration time. The study suggests the feasibility of using amino acid in the development of zero saccharide rapid disintegrating tablets (RDTs) by lyophilisation.
