High-throughput antibody display methods are useful for identifying novel variants from large sequence landscapes but are often limited to screening on binding properties alone. Here we present a novel platform for rapid antibody discovery based on binding and functional activity readouts. Our microcapillary array technology, which is essentially up to a 20 million well microtiter plate, allows us to screen mammalian, yeast, or bacteria cells or their protein products in a wide variety of assay formats, including reporter cell stimulation, cross-reactivity screening, and cell surface binding assays. This novel hardware and software platform combined with our proprietary human inspired synthetic antibody library has enabled the isolation of antibodies against multiple clinically relevant targets.
