In developing protein drugs, the reduction of production cost by increasing protein expression is very important for the competitiveness of protein drugs. In response, most companies are now working on the development of new expression tools such as expression vector improvement and cell culture media improvement to increase secreted protein expression. Signal peptide could be a good tool to improve secreted protein expression. Signal peptide is well-known to play a very important role in protein secretion. In addition, the signal peptide plays a role in the expression of secreted proteins. The signal peptide can determine the amount of secreted protein in concert with the type of secreted protein. At present, it seems like to be about 6,500 signal peptides present. These signal peptides greatly affect the expression level of each secreted protein. Here we prepared more than 1,200 signal peptides and examined the expression of various proteins following them. The re suits show that the amount of protein secreted is determined not by the signal peptide but by the compatibility of the signal peptide with the following protein.
