Directed evolution of enzymes by sequential cycles of random mutagenesis and screening have proved to be useful for obtaining new or improved properties. This strategy is thought to be coventient and even primitive as no structural infomation is required. Theoretical works on fitness landscapes is also underway with the same aspects and motivations. Therefore, it may be best to integrate both experimental and theoretical aspects so as to provide a more efficient strategy for the adaptive walk in directed evolution. To construct an efficient strategy of adaptive walk in directed evolution, it is essential to know the folowing basic parameters characterizing a random mutant population: average and variance of the effects of one-point mutations and degree of nonadditivity of the mutational effects.
