Hyperuricemia is a condition characterized by levels of blood's uric acid that is higher than its normal levels. If left unchecked, hyperurecemia will trigger gout, an accumulation of mononosodium urate crystals caused by a disruption of purine metabolism in the joints, periarticular tissues, bones and other organs. Xantine oxidase (XO) is an enzyme that plays a role in the reaction of the hypoxantine oxidation reaction to xantine and becomes uric acid. Therefore, inhibition of XO activity becomes the main focus in the treatment of gout. Some medicinal plants are known to inhibit XO activity. Water extract of tempuyung (Sonchus arvensis) has been studied to be able to inhibit XO activity and potentially be developed into a gout medicine. In this study fractionation of tempuyung water extract was conducted to look for compounds that play an active role as XO enzyme inhibitors. The electrochemical method was used to test the inhibition of XO of the tempuyung's extract and fractions. Tempuyung water extract was fractionated using liquid-liquid extraction and obtained 4 fractions, namely n-hexane fraction, dichloromethane fraction (DCM), BuOH fraction and water fraction. The XO inhibition activity test showed that all active fractions inhibited XO, with the DCM fraction showing the highest inhibition agains XO activity, with IC_(50) of 119.02 ppm.
