Oligonucleotides containing 2'-pyrene modified uridine(Upy) exhibit the increase of pyrene monomer emission whenhybridize with the complementary RNA).The increase offluorescence intensity is caused by the location of pyrene at theminor groove of A-form duplex.To examine the effect of restrictedC3'-endo conformation of Upy on the fluorescence intensity ofpyrene at duplex,a DNA containing pyrene-modified amide-linkeddinucleosides (UpyadODN:5'-dCATGUpyadCTAC-3') wassynthesized and it showed the increase of fluorescence intensity to7-fold at 375 nm when hybridized with RNA).However,theincrease of fluorescence intensity was smaller than that of duplexwhich consists of DNA containing Upy and the complementary RNA.To enhance thefluorescence intensity of the oligonucleotide containing pyrene-modified amide-linkeddinucleosides when it hybridizes with RNA,we designed a 2'-O-methyl RNA containingamide-linked RNA modified with pyrene at the 2'-position (C~(OMe)A~(OMe)U~(OMe)G~(OMe)UpyadU~(OMe)U~(OMe)A~(OMe)C~(OMe),1,Figure 1).Here we report the solid-phase synthesis of 1.
