Use of an autogenous PRRSV vaccine in conjunction with MLV elicits broadly neutralizing antibodies

Porcine reproductive and respiratory syndrome virus (PRRS V) i among the most important viruses impacting the swine industry on a worldwide basis.1 Since its widespread emergence in the late 1980s the virus has evolved rapidly resulting in tremendousgenetic diversity.2 Most recently, a new PRRSV strain, identified by RFLP pattern "1-7-4" has emerged as a significant threat to the health of both the growing-pig and breeding herds in the United States.3 Constant antigenic drift within circulating strains affects vaccine efficacy and hinders the development of new vaccines in a timely manner. Commercial modified live virus (MLV) PRRSV vaccines can offer homologous protection. However, they often exhibit reduced efficacy when used to combat disease caused by heterologous strains circulating in the field.4 Inactivated autogenous vaccines can be manufactured to match the outbreak or circulating field strain, however, their efficacy has been debated.4 The objective of this study was to broaden the antibody repertoire by using modified live vaccines in combination with a PCV2 vaccine and a bivalent autogenous PRRSV vaccine in a prime-boost strategy. The results demonstrate that the inactivated vaccine was able to elicit neutralizing antibodies with broader specificity than those made in response to the MLV vaccine alone.
