This paper investigated the genetic variations of the strains of the species Auricularia at the molecular level in order to protect Auricularia resources from reducing and provide a theoretical basis for the breeding of new varieties and laid a foundation for the construction of the DNA fingerprint of Auricularia. This research used ISSR markers to assess the germplasm genetic diversity among 55 individuals of Auricularia. 17 primers were selected from 27 primers for germplasm analysis. Our results showed that the 17 primers amplified 155 fragments, and the polymorphic loci were 155 and the percentage of polymorphic loci reached 100%. The Jaccard's genetic similarity matrix and dendrogram for the 55 strains were formed based on the ISSR-generated bands. In dendrogram, the 55 strains could be divided into five groups, which contained 12, 11, 2, 24, and 6 strains, respectively. The average genetic similarity coefficient of the 55 strains was 0.4109.
