A practical method was developed for the analysis of 20 N-(O,O-diisopropyl) phosphoryl amino acids (Dipp-aas) by HPLC/UV/ESI-MS with an NH_2 column (150mmx4.6mm i.d). The column was eluted with a mobile phase of ACN/10 mM NH4Ac (10:90, v/v, pH 7.5) at a flow of 0.8 mL/min. A good linear correlation (R~2>0.8) between the estimated partition coefficient (n-Octanol/Water) Log P and the Log k' of Dipp-aas on NH_2 column was observed. This correlation equation could be applied to predict the retention mechanism of amino acids and Dipp-aas. Meanwhile, the developed method was successfully used for the investigations of purity and stability of Dipp-aas in the given mobile solution. The potential decomposition substance of Dipp-aas were characterized by ESI-MS.
