SINEUPs: Novel Antisense ncRNA Can Enhance Protein Translation

Mouse Uchl1 gene is sharing anti-sense ncRNA as 5' prime head to head at chromosome 5. In the Parkinson's disease, UCHL1 (PARK5) expression is suppressed because of mutation of uchl1 gene. Our collaborator Gustincich found AS ncRNA which is including inverted SINEB2 can up-regulate UCHL1 protein expression but not mRNA after rapamycin treatment. Interestingly, ASGFP which is modified sharing part can also up-regulate GFP protein expression level. We have known that Inverted SINEB2 in AS ncRNA is important to protein up-regulation. If we can create small molecule of protein up-regulater, it has potential to produce many industrial protein or therapeutic target protein. I will show how to define the key region of AS RNA and how to screen key structure of InvSINEB2.
