Protein-based biologies have been increasingly highlighted as promising therapeutics because of their potential for enhanced target binding, reduced toxicity, and improved pharmacological properties compared to small molecule drugs. Due to the vast diversity of amino acid sequence space and the limited ability to rationally design proteins with desired biological outputs, developing a protein-based therapeutics generally requires sampling of a large number of variants. We have developed a high-throughput platform called muSCALE (Microcapillary Single Cell Analysis and Laser Extraction) for rapid evaluation of millions to billions of protein variants produced from yeast or bacteria.
