To investigate the frequency-properties of propagation of local field potentials (LFPs), we developed a combination method of optogenetics and micro-electrocorticogram (micro-ECoG). Photo-stimulation of the deep layer excitatory cells in the rat primary visual cortex evoked widely distributed ECoGs responses and intracortical LFPs. Comparing their response-to-frequency slopes, low-pass property of vertical LFP propagation and less frequency-dependent LFP propagation in horizontal directions were suggested. The vertical LFP propagation also involved intrinsic gamma-band power generation. The combination of optogenetics and micro-ECoG provides a hopeful solution to wide-areal, time-precise investigation of cortical functions.
