Shizpphyllan (SPG), a kind of β-1,3-glucan, is known to be recognized by dectin-1 on antigen presenting cells. It was also revealed that SPG can form a complex with polynucleotide, and this complex consists of two polysaecharide-strands and one polynuleotide-strand. The complex consisting of SPG and oligonucleotides was prepared and the formation was confirmed by gel permeation chromatography. Furthermore, the complex could inhibit the binding of biotin-labeled SPG to the cells having dectin-1. These data indicate that SPG can be recognized after the complexation with oligonuicleotides (ODNs) and has an ability to deliver ODNs into antigen presenting cells. These finding indicate that β-1,3-glucans are very attractive and useful materials in gene delivery technology.
