A three-dimensional (3D) hyaluronic acid (HA) and gelatin (Gtn) fibrous scaffold was successfully fabricated to mimic the architecture of natural extracellular matrix (ECM) based on electrospinning. Thiolated HA and Gtn derivatives, HA-DTPH and Gtn-DTPH, were synthesized and electrospun to form composite 3D fibrous scaffolds. In order to facilitate the fiber formation during electrospinning, Poly (ethylene oxide) (PEO) was added into the aqueous solution of HA-DTPH and Gtn-DTPH at an optimal weight ratio of 1:1.2. The electrospun HA-Gtn/PEO blend scaffold was subsequently cross-linked through Poly (ethylene glycol)-diacrylate (PEGDA) mediated conjugate addition. PEO was then extracted in deionized (DI) water to obtain an electrospun HA-Gtn fibrous scaffold. Adult human dermal fibroblasts (AHDFs) were seeded on HA-Gtn fibrous scaffold for 24h in vitro. Laser scanning confocal microscopy (LSCM) revealed that the AHDFs attached to the scaffold and spread, which suggests potential applications of HA-Gtn firbous scaffolds in tissue regeneration.
