Potentiometric sensors allow for point-of-care serological biosensing, with comparable results to SPR and ELISA. A thiol-based self-assembled monolayer (SAM) allows for covalent functionalization of antigens, and, therefore, selective detection of antibodies in solution. The carboxyl-SAMs allow large degrees of non-selective adsorption to the sensor surface. Pre-blocking is a promising method to block nonspecific sites without using high concentrations of designer receptor molecules. Pre-blocking efficiency will be demonstrated with potentiometric measurements, and parameters to maximize covalent functionalization density will be explored.
