摘要

The analysis of macromolecules from 100 kDa to Mega-Dalton masses is a new frontier in mass spectrometry (MS). Macro-ions result in low signal levels at high m/z due to poor signal responses from conventional detectors. Solutions to this detector problem have involved increasing the charge-state of ions by electrospray ionization so that a high ion velocity is maintained or using a detector that can respond to low velocity, low charge-state ions. Described here is the use of a cryodetector based MALDI TOF mass spectrometer that can respond with high sensitivity to heavy ions from polystyrene at 2 MDa, PMMA at 253 kDa and IgG at 150 kDa.
