Development, Characterization and Application of a UV-Sensitive Bacteriophage for UV Reactor Challenges

A new microorganism suitable for UV reactor validation was isolated from an environmental source and characterized with respect to its morphology, culturing procedure, and sensitivity to the UV radiation. Itwas named T1UV, due to its DNA similarity to the T1 bacteriophage group. The UV dose-response at 253.7 nm of T1UV phage followed first-order kinetics with a calculated D10 value of approximately 5 mJ cm?2, making it a well suited test surrogate for Cryptosporidium parvum and Giardia lamblia. T1UV is grown using an E. coli CN13 host at titres as high as 7×10~(10) PFU/mL, and is thus compatible for use in tests in combination with other common challenge bacteriophage that are grown using different hosts (MS2, Q-Beta). UV spectral sensitivities of T1UV and Q-beta were determined at selected wavelengths from the UV emission spectrum of medium pressure lamp. The UV action spectra of the two microorganisms are similar to that of MS2, and different from the DNA absorption spectrum.
