NMR Investigation of Magnesium Specific Binding to DNA Duplex

This work deals with interaction of DNA and magnesium ions. Two self-complementary DNA oligonucleotides, namely 5 '-GATGCATC-3' and 5 '-CTTGCAAC-3 ', were employed as molecular models of DNA duplex. Their high resolution ~1H and ~(31)P NMR spectra were measured as a function of magnesium concentration in aqueous solution. It has been released that at a room temperature, when the oligonucleotides are present in a form of stable duplex, the influence of increased magnesium content on chemical shifts is remarkable for numerous nucleobase protons as well as for phosphorus nuclei of the phosphate groups. The effect depends on the position of the particular nucleotide in the DNA chain, which indicates at least partial local magnesium binding.
