The synthesis of polymer-protein conjugates for their application as therapeutics is a rapidly expanding area of research. The main drawbacks associated with current protein therapeutics are their susceptibility towards proteolytic degradation and rapid renal excretion in vivo. Pioneering work by Abuchowski et alhas demonstrated that the covalent attachment of polymers to protein therapeutics can give rise to several beneficial effects. Poly(ethylene glycol) (PEG) is by far the most commonly used polymer for protein conjugation due to its ease of functionalization, low cost and water solubility.
