Synthesis of Aminooxy End-Functionalized Polymers by RAFT Polymerization for Bioconjugate Formation

Controlled radical polymerization techniques have been used in recent years to prepare a wide variety of well-defined polymers. Among the methods employed is reversible addition-fragmentation chain transfer (RAFT) polymerization. This robust technique has proven to be tolerant of a wide range of monomers. Furthermore, functional groups can be incorporated at the chain ends by appropriate design of the chain transfer agent (CTA). Herein, we report the synthesis of an aminooxy-functionalized CTA. We utilized this CTA to produce ‘smart’ polymers. In particular, the thermosensitive poly(N-isopropylacrylamide) (pNIPAAm) was synthesized. Oxime bond formation is an excellent method to prepare protein-polymer conjugates, and we plan to utilize this method to synthesize polymers for well-defined bioconjugate formation.
