We have been focusing much of our attention on the synthesis and biological testing of metal-containing polymers as part of a battle against cancer -22 More recently, we started to look at the ability of these materials to inhibit viruses 4-18 Much of this effort involved the use of polymeric derivatives of cisplatin and organotin-containing polymers. We employ Balb 3T3 cells as our initial target organism to investigate a material's biological activity. Past studies have shown that results derived from testing Balb 3T3 cells are good indicators for a polymer's ability to inhibit actively growing cells. Balb 3T3 cells are also relatively easy and inexpensive to maintain. Many of the compounds synthesized and tested contained known antibacterial and antiviral drugs such as norfloxacin, ampicillin, ciprofloxacin, and acyclovir. In general, the ability to inhibit cell growth, with respect to the alkyltins is Bu>Pr>Et>Me>Oc>La with the methyl, octyl, and lauryl all essentially inactive. Our overall rationale for employing drugs coupled with organotin moieties with known biological effects is to create a compound that can interact with the target (cancer, bacteria, virus) at several venues, increasing the effectiveness of the combination drug against the target. Recently, we found that several simple non-drug containing polymers showed good inhibition of Balb 3T3 and human ovarian adenocarcinoma cells (Caov-3). One of these compounds was the polymer made from dibutyltin dichoride and 2- chloro-1,4-benzenediamine that showed a GI50 value of 0.04 μg/mL against Balb 3T3 cells.
