Detection of proteins is needed in many situations. In many cases, not only high sensitivity but also portability is required. To this end, electrochemical principles and devices are advantageous. We have already reported a sensing device based on coulometric detection of an enzymatic reaction product (H_2O_2) in a solution plug coupled with silver metallization [1]. By separating the oxidation and reduction reactions in two flow channels to improve collection efficiency and reduce background, significant enhancement of sensitivity and lowering the detection limit was demonstrated. We demonstrate the application of this technique to the detection of proteins based on enzyme-linked immunosorbent assay (ELISA).
