Effects of different initial sucrose concentration were compared on the cell growth, anthocyanin production and other nutrients usage in Hibiscus sabdriffa L. suspension culture. Cell growth increased along with the increase of initial sucrose concentration below 40 g/L, 10 g/L sucrose became the limiting factor to facilitate a linear growth. While a initial concentration higher than 40g/L became stressful for cell growth characterized by an extend lag phase, but provoked a metabolsim shift favoring the anthocyanin synthesis and accumulation. Browning of the medium and premature cell death was also observed. Maximum cell density, anthocyanin content and total anthocyanin amount were 14.36 gDW/L, 21.8 mg/gDW, and 260 mg/L, at initial sucrose concentration of 40, 60, 40 g/L. Sucrose was hydrolyzed into hexoses, which were taken up and metabolized. Nitrate shows more importance on balancing the nutrient composition to keep a stable ratio of introgen to carbon. Other trace nutrients, ammonium and phosphate, were uptaken quickly and accumulated in the cell to regulate cellular metabolism.
