The saccharide primer method was developed as a novel method for oligosaccharide synthesis. The saccharide primer such as 12-azidododecyl β-lactoside, supplemented in culture medium, is administered into cells and glycosylated by cellular glycosyltransferases in Golgi to afford pseudo-glycolipids, which are secreted into the culture medium. By the sacchride primer method, various kind of oligosaccharides can be synthesized efficiently depending on the cell's own characteristics and oligosaccharide synthetic ability. The oligosaccharides bearing the azido group can be obtained easily by introducing azido group into the saccharide primer. We have reported that tetraphenylethylene (TPE) compounds bearing 6'-sialyllactosyl moieties (α2,6SL-TPE) were synthesized and used for detection of influenza virus[1]. In this study, we aimed to evaluate oligosaccharide-binding affinity of macromolecules such as receptor proteins expressed on cell surface and target molecules bound on microbeads by using flow cytometer.
