We report that oligonucleotides with specific end-modification within a plug in a plug-based microfluidic device to undergo a locally enhanced concentration at the rear of the plug as the plug moves downstream, based on a combinatorial effect involving hydrodynamic trapping provoked by internal vortices, and interfacial adsorption caused by affinity attraction. The results demonstrate that rapid DNA hybridization is achieved in the device in seconds (~3.8 s), and the enrichment of local concentration occurs in a nL~pL free-solution plug, thus profiting the DNA detection.
