Quantitative Analysis of Total Doxorubicin, Free Doxorubicin and Doxorubicinol in Human Plasma with LC-MS/MS

A rapid and simple LC-MS/MS method to quantify free DXR, total DXR and DXR-ol in human plasma was developed and validated over the curve range of 0.500-500 ng/mL (free DXR and DXR-ol) and 1.00-1000 ng/mL (total DXR). No significant (<5percent) release of liposomal DXR was observed during SPE extraction. Liposomal DXR can survive at least three freeze-thaw cycle and is stable for at least one month under -20 deg C.
