1. This method demonstrates a workflow of novel MS-based screening of biomarkers from discovery to qualification. The use of MRM for biomarker qualification conveys a number of advantages, allowing the detection of multiple peptide biomarkers in a short period of time. 2. An ideal biomarker candidate for K. pneumoniae should be one specific to K~(+) and common in all K~(+) patients. No peptides were selected based on current results. Negative results might be due to extremely low abundance of peptides in urine, false positive results from shot-gun sequencing, peptide instability in patient samples, matrix interference of patient samples, etc. Follow-up studies include screening from plasma, enrichment of urine samples using antibody approaches, or qualification with orthogonal approaches such as ELISA.
