Hepoxilins(Hx) are epoxy alcohols formed through the 12-lipoxygenase pathway in various animal cells. In human cells synthesis of hepoxilins is catalysed by the enzyme 15-lipoxygenase type 1 (15-LO-1), leading to the formation of 14,15-hepoxilins (14,15-Hx). 15-LO-1 inserts molecular oxygen to arachidonic acid leading to the formation of the unstable intermediate 15-hydroperoxyeicosatetraenoic acid (15-HPETE) which can be further metabolized. Here we show that the human Hodgkin cell line L1236 which possesses high 15-LO-1 activity transforms arachidonic acid to cysteinyl-containing 14,15-Hx. These cysteinyl-containing 14,15-Hx isomers has not earlier been identified in human cells.
