A simple analytical method based on MDMS-SL strategy was established to identify and quantify DAG species present in biological samples. Facile derivatization of DAG species with DMG was exploited in the developing method. This derivatization procedure led to enhancing their ionization by ESI-MS, and increasing the specificity with a unique set of fragment ions. This method possessed high reproducibility (<8%), nearly 4 orders of linear dynamic range and a limit of quantification at lower than 100 amol/μL. Moreover, this method enabled us to discriminate between sn-1,2- and 1,3-DAG isomers. We believe that development of this new method could facilitate identifying the importance of the isomers in lipid metabolism and biological functions.
