LC-MS/MS (2hrs) analyses of hipocampal proteome extracted from rats, which were subjected to methamphetamine treatment, permitted the identification of 1829 unique peptides corresponding to 302 proteins. 11 proteins exhibited a significant up- or down-regulations as suggested by their spectral counts. These changes were also confirmed by MRM experiments. Synaptic vesicle glycoprotein 2A (p= 0.0001 and +18%) exhibited the most significant up-regulation while Spectrin alpha chain non-erythrocytic 1 (p= 0.0002 and -26%). Synaptic vesicle glycoprotein 2A is involved in the control of regulated secretion in neural and endocrine cells, and enhancing selectively low-frequency neurotransmission. Spectrin alpha chain non-erythrocytic 1 is involved in secretion, interacts with calmodulin in a calcium-dependent manner. LC-MS/MS (2hrs) analyses of olfactory bulb proteome extracted from rats, which were subjected to methamphetamine treatment, permitted the identification of 1975 unique peptides corresponding to 336 proteins. 7 proteins exhibited a significant up- or down-regulations as suggested by their spectral counts. These changes were also confirmed by MRM experiments. Guanine nucleotide-binding protein G(i) subunit alpha-1 (p= 0.001 and +18%) exhibited the most significant up-regulation while Alpha-2-HS-glycoprotein (p= 0.0005 and -20%) exhibited the most significant down-regulation. Guanine nucleotide-binding protein G(i) subunit alpha-1 is involved as modulators or transducers in various transmembrane signaling systems. Alpha-2-HS-glycoprotein is involved in inhibiting both insulin-receptor tyrosine kinase activity and insulin-stimulated receptor autophosphorylation. The abovementioned protein expression up- and down-regulation are involved in many biological pathways and are interrelated.
