High sensitivity and simultaneous quantitation method for arginine vasopressin and desmopressin in human plasma determined by LC-MS/MS/MS

The calibration curve for AVP showed the linearity in a range between 0.2 and 80 pmol/L. The calibration curve for DDAVP showed the linearity in a range between 2 and 800 pmol/L. The calibration range and the accuracy (±15.0% and lower) were acceptable, respectively in both calibration curves. In addition, this gradient mode of HPLC condition was possible to separate AVP and DDAVP completely. It was able to use a scheduled MS/MS/MS acquisition, and it was improved sensitivity for AVP and DDAVP. It was suggested that our method was very useful to detect AVP and DDAVP levels in human plasma collected from 25 volunteers. The parameter "R" squared (R~2) of linear regression between the quantitation results by LC-MS/MS/MS and RIA was 0.9213. This LC-MS/MS/MS method has the potential to supplant the RIA.
