The benefits of using ice as a matrix to improve signal abundance in IR-MALDESI tissue imaging was confirmed and supported by coupling shadowgraphy and MS data acquisition. The effect of spotto- spot distance, number of pulses per pixel and several other parameters were systematically investigated and up to 10 folds improvement in signal was achieved.
