Tandem mass spectrometry conclusively identifies the HMWC of spirochete T. denticola as composed exclusively of FlgE. Tandem mass spectrometry suggests peptide T15 is important to the cross-linked HMWC species. Tandem mass spectrometry identifies a likely cross-linked peptide species from T13/14 and T15 at m/z 3731.9397 u. Mutation of selected amino acids on T15 suggests the roles of Asp~(181), Asn~(179) and Cys~(178) in the biochemical mechanism of isopeptide bond formation and covalent cross-linking of FlgE in the HMWC.
