SCX SPE efficiently removed most of the background peptides and therefore reduced matrix effect and background noise level. SCX SPE improved the sensitivity of peptides VVSV and SLIY. SCX SPE was more efficient than RP SPE for the removal of the abundant background peptides. 2D (RP+SCX) SPE did not significantly improve the extraction cleanness. A sensitive, accurate and reproducible LC-MS/MS assay was successfully developed to quantify BMS-986012 in monkey serum using SCX SPE.
